As mentioned in the previous article, CytoNiche microcarrier (V Series) can be used to culture a variety of adherent cells for the production of viral biological products. How about its actual virus production performance?

Vero Cells (African Green Monkey Kidney Cells)

Vero cells have the following application advantages:

1. Easy to establish cell bank, easy to preserve, fast growth rate and can obtain continuo us cell line;

2. Stable genetic characters, non-tumorigenic in a certain passage, and excellent biosafety;

3. Sensitive to a variety of viruses, high virus titer;

4. Defect in type I interferon secretion, and more vulnerable to virus infection;

5. High attachment ability, well attached to the surface of microcarrier, and easy to achieve scale-up culture.

Therefore, Vero cells are widely used in a variety of vaccine production, such as Encephalitis B, Hemorrhagic Fever with Renal Syndrome, Rabies, Poliomyelitis, Hand-Foot-Mouth Disease, Rotavirus, etc.; In addition, Vero cells also show good performance in the emerging field of oncolytic viruses.

CytoNiche microcarrier enables continuous scale-up subculture of Vero cells in a multi-stage reactor, and the peak cell density can reach 1×107/mL. It is easy to achieve process scale-up. Using CytoNiche microcarrier to culture Vero cells and harvest the viruses, the titer of virus A is nearly 2 orders of magnitude higher than that of traditional 2D technology.

Compared to the traditional microcarrier, the virus B and virus C produced by Vero cells were cultured using the traditional microcarrier, and the efficiency of virus production is also significantly higher than the conventional microcarrier.

  

MDCK Cells (Dog-Renal Epithelial Cells)

MDCK cells have the following application advantages:

1. Easy to establish cell bank, easy to preserve, fast growth rate and can obtain continuous cell line;

2. Sensitive to a variety of viruses, short of toxic cycles, high virus titers;

3. After continuous passage in MDCK cells, the virus can still maintain high antigen stability.

MDCK cells are sensitive to a variety of influenza virus subtypes, and are widely used in the production of multivhetic flu vaccines.

CytoNiche microcarrier enables continuous scale-up subculture of MDCK cells in a multi-stage reactor, and the peak cell density can reach 1×107/mL before seeding viruses. Using CytoNiche microcarrier to culture MDCK cells and harvest the viruses, the hemagglutination titer of virus D is 4 times that of traditional spherical microcarrier.

Human Diploid Cells

Human diploid cells have the following application advantages:

1. No potential carcinogenicity, excellent safety;

2. Susceptible to a variety of viruses, suitable for the production of a variety of viruses;

3. No animal source protein, factor pollution;

4. The inactivated virus produced in diploid cells features high immunogenicity, fast immune

Human diploid cells are widely used in the production of inactivated vaccines such as Hepatitis A, Chickenpox, Poliomyelitis and Rabies virus, as well as Live Attenuated Rubella vaccines; In addition, they are also used in the production of Fibroblasts Interferon and other products.

Using CytoNiche microcarrier to culture human diploid cell line, when harvesting, the number of cells per gram of microcarrier is 3–4 times that of traditional microcarrier, and the cell density can reach 2×106/mL. Among them, using CytoNiche microcarrier to culture type I human diploid cells and harvest the viruses, the antigen content of virus E is about 3 times that of 2D process.

 

The 3D dissolvable porous microcarrier independently developed by CytoNiche can be used to culture a variety of adherent cells for the production of viral biological products. Compared with traditional microcarrier, it has significant advantages in cell expansion and virus production.
