In recent years, there are more and more research and development projects in the fields of oncolytic viruses, viral vectors, and novel vaccines. Many of these emerging viral biological products are relying on adherent cell culture for virus production. With the traditional culture process, adherent cells are usually cultured in cell culture flasks, roller bottle, or cell factories. The culture scale is relatively limited, the culture process is difficult to control, and it is difficult to scale up the process under the premise of ensuring quality. Using microcarrier in combination with bioreactor for suspension culture of adherent cells is a very good solution.

In most cases, traditional microcarrier are rigid solid microsphere prepared with synthetic polymers, and the adherent cells grow in a 2D curved pattern on the surface of microcarrier. At high feeding density, stirring in the reactor is prone to cause cell death and generate microcarrier fragments, which can lead to cost increase, safety risk, and low recovery rate in virus production and subsequent purification step.

CytoNiche launched the first 3D dissolvable porous microcarrier—3D TableTrix® Microcarriers (V Series) for the industry of viral biological products. It can be used not only for the breeding, harvesting, and subculturing of adherent cells, but also for virus production; Compared with the traditional microcarrier, it has significant advantages in cell expansion and virus production.

Features

1. Honeycomb porous structure, pore size range 30–50 μm, porosity > 90%;

2. Size range of 50~500μm can be customized;

3. The specific surface area is more than 9000cm2/g (about 2–3 times the surface area of traditional microcarrier), which can provide more growth space and hold more adherent cells with the same feeding density;

4. It features bionic biomechanical characteristics, which can effectively reduce the fluid shear force and mitigate collision between microcarrier in the stirred reactor, and improve the cell viability, so as to improve the viral yield;

5. The microcarrier can be completely dissolvable, a microcarrier degradation residue detection method (kit) verified by the National Institutes for Food and Drug Control is available.

 

 

Qualification

 Two core microcarrier products developed by CytoNiche have passed the review, approval, and filing of CDE (registration number: F20210000003, F20210000496)

√ "Gelatin Microcarrier Tablet for Cells*" has completed the filing of the FDA DMF qualification for pharmaceutical excipients (DMF: 35481, activated). It is the only microcarrier product in the DMF list announced on the official website of FDA.
Gelatin Microcarrier Tablet for Cells*: Spherical microcarrier are preformulated and packaged as tablets.

The PECALS ™ system is compatible with 3D FloTrix® CELL PRO viable cell on-line monitoring system, and can precisely monitor the status of viable cells in real time and automatically generate cell proliferation curve, making cell culture easier!

Combined with CytoNiche Microcarrier and 3D FloTrix® vivaSPIN Automation Bioreactors, the Vero cell culture can achieve a high density of 1×107/mL.
